A 25 mT Preclinical Electron Paramagnetic Resonance Oxygen Imager, JIVA-25, And Its Applications to Biomedical Studies

Abstract: Local hypoxia, or low oxygenation, is an important factor affecting tumor and islet transplantation therapies for type I diabetes cure. Biomedical scientists studying tumor, T1D, and many other biomedical fields need a noninvasive method to map tissue partial oxygen pressure (pO2). Electron paramagnetic resonance oxygen imaging (EPROI) is emerging as a leading technology that can provide accurate pO2 maps in vitro and in vivo. O2M developed the world’s first dedicated preclinical oxygen imager, JIVA-25, based on EPROI. JIVA-25, provides three-dimensional pO2 maps with high spatial, temporal, and pO2 resolution. For reporting oxygen concentration, JIVA-25 uses oxygen-dependent relaxation rates of trityl radicals OX063 or its deuterated version OX071. During the past two years, JIVA-25 was used to perform (a) oxygen guided radiation therapy study on SCVII tumor model, (b) combination study with a radiosensitizer and radiation therapy on primary sarcoma and E0771 tumor models, and (c) In vitro and in vivo pO2 assessment of several islet encapsulation devices as part of the JDRF funded “Oxygen Measurement Core” project. I will present key data from these studies and discuss the JIVA-25 instrument capabilities.
